Allergen Potential of Timothy Grass Aerosols by Immunogold Electron Microscopy

Filtration of biological hazardous materials like allergen active particles form the indoor environment could minimize the health risk of allergy sufferers. To classify air filters relating their filter potential of bioactive particles, a detailed knowledge of present aerosols is prerequisite. To this aim an experimental setup has to be established, wherewith a controlled growth environment of an allergy potential plant can be realized, exemplary timothy grass (Phleum pretense). Immunogold labelling against the major allergen under investigation (Phl p5) will reveal at which grow state the allergen is already present in the aerosol released by the plant.

The aim of this thesis is to reveal detailed knowledge about the present and spatial distribution of allergens present at timothy grass aerosols, except pollen grains themselves.

  • Construction of experimental setup for controlled timothy grass germination
  • Immunogold labelling protocol adaptation for given samples
  • Sample collection and analysis using Environmental Scanning Electron Microscopy (ESEM)
